Which GPU for what purpose?

Is there a benefit if I assign a GPU for OpenCL and the other one for CUDA?

or should I just leave it at default where both options have GPU #1 assigned ?

There's no benefit to keeping them different, no. You should choose the most powerful GPU available from the list for both options.
